If you got a puter that has plenty of slots and you are going to install a total of 12 gig of memory.
 
Is there a advantage of using three 4g vs six 2g memory cards and if so is it worth it?

Not exactly.  Some motherboard/processor combinations only support dual channel RAM, which works best in matched pairs.  This is old enough that it probably is dual-channel, and that would make the 3 x 4 GB option less effective.
